How does Longifene work?

Longifene contains buclizine. It exerts its anti-emetic effect by binding to and blocking the muscarinic and histamine receptors in the vomiting center of the central nervous system (CNS). This prevents the activation of the chemoreceptor trigger zone (CTZ) and may reduce nausea and vomiting. The vomiting center possesses neurons which are rich in muscarinic cholinergic and histamine-containing synapses. Motion sickness principally involves the overstimulation of these pathways due to various sensory stimuli. Longifene blocks the histamine receptors in the brain (vomiting center) and thus reduce the activity of these pathways which is responsible for motion sickness. In this way Longifene provides relief to the patient from symptoms associated with motion sickness.
