Rafinlar 75mg Capsule

Rafinlar, containing Dabrafenib, is a targeted therapy drug used in the treatment of certain cancers with BRAF V600E or V600K mutations. It is a kinase inhibitor that blocks abnormal cell growth and division, offering a precision medicine approach for patients with these specific mutations.

Uses

• Unresectable or metastatic melanoma with BRAF V600E or V600K mutations.
• Metastatic non-small cell lung cancer (NSCLC) with BRAF V600E mutation (in combination with trametinib).
• Advanced or metastatic anaplastic thyroid cancer (ATC) with BRAF V600E mutation (in combination with trametinib).

Dabrafenib inhibits the activity of mutated BRAF kinase, which is part of the MAPK/ERK signaling pathway. By blocking this pathway, it prevents cancer cells from growing and dividing uncontrollably.

Side Effects
Common side effects :

• Fever and chills
• Nausea and vomiting
• Fatigue
• Skin rash or irritation
• Joint or muscle pain

Severe side effects:

• Secondary skin cancers
• Hyperglycemia
• Heart problems (e.g., decreased left ventricular ejection fraction)

Precautions

• Test for BRAF V600E or V600K mutations before initiating therapy.
• Monitor for signs of skin changes and secondary cancers.
• Regularly check blood glucose levels in diabetic patients.
• Avoid in pregnancy and breastfeeding due to potential harm to the fetus or infant.

Dosage and Administration

Formulation: Capsules for oral use

Recommended Dosage:

• Typically 150 mg orally twice daily, taken on an empty stomach (at least 1 hour before or 2 hours after a meal).

Combination Therapy: Often used with trametinib for enhanced efficacy.
