Lenvima, containing the active ingredient Lenvatinib, is a potent oral tyrosine kinase inhibitor (TKI) used primarily for the treatment of various cancers. It works by blocking specific proteins involved in tumor growth and angiogenesis.
Thyroid Cancer: Treatment of radioactive iodine-refractory differentiated thyroid cancer (DTC).
Renal Cell Carcinoma (RCC): Used in combination with everolimus for advanced RCC after one prior anti-angiogenic therapy.
Hepatocellular Carcinoma (HCC): First-line treatment for patients with unresectable HCC.
Endometrial Carcinoma: Used in combination with pembrolizumab for advanced endometrial carcinoma that is not microsatellite instability-high (MSI-H) or mismatch repair deficient (dMMR).
Lenvatinib inhibits multiple receptor tyrosine kinases (RTKs), including vascular endothelial growth factor receptors (VEGFR1, VEGFR2, VEGFR3), fibroblast growth factor receptors (FGFR1-4), and other pathways that promote cancer cell proliferation and angiogenesis.
Onset: Effects on tumor growth can vary depending on the cancer type and patient response.
Duration: Typically requires continuous treatment as prescribed.
Thyroid Cancer: 24 mg once daily until disease progression or unacceptable toxicity.
Renal Cell Carcinoma: 18 mg in combination with 5 mg everolimus once daily.
Hepatocellular Carcinoma: 8 mg for patients <60 kg, 12 mg for patients ≥60 kg, once daily.
Endometrial Carcinoma: 20 mg once daily in combination with pembrolizumab.
