Tybriva Information

Tybriva contains Ibrutinib, a targeted therapy medication used for treating various types of blood cancers. It belongs to a class of drugs called Bruton’s tyrosine kinase (BTK) inhibitors. By blocking BTK, Ibrutinib interferes with cancer cell survival and proliferation.

Uses

  • Chronic Lymphocytic Leukemia (CLL)
  • Mantle Cell Lymphoma (MCL)
  • Waldenström’s Macroglobulinemia (WM)
  • Marginal Zone Lymphoma (MZL)
  • Chronic Graft-Versus-Host Disease (cGVHD)

Mechanism of Action

Ibrutinib irreversibly binds to Bruton’s tyrosine kinase (BTK), inhibiting B-cell receptor signaling and reducing the survival and proliferation of malignant B cells.

Onset of Action

Clinical improvements are often observed within a few weeks of starting treatment, though full effects can take longer.

Duration of Action

The half-life of Ibrutinib is approximately 4 to 6 hours, but its therapeutic effects can last much longer due to irreversible BTK binding.

Dosage

  • CLL and WM: 420 mg orally once daily.
  • MCL: 560 mg orally once daily.
  • cGVHD: 420 mg orally once daily. The dosage should be adjusted based on patient-specific factors and medical supervision.

Side Effects

  • Diarrhea
  • Fatigue
  • Muscle and joint pain
  • Bruising or bleeding
  • Hypertension
  • Infections

Precautions

  • Monitor for bleeding, as Ibrutinib can increase the risk of hemorrhage.
  • Regularly check blood pressure during treatment.
  • Avoid use in patients with severe liver impairment.
  • Use effective contraception during treatment and for some time after stopping.
